If my final sequencing will be on PacBio, does QC on the Plasmidsaurus ONT device still reflect performance?

Yes. EpiCypher routinely uses Plasmidsaurus Fiber-seq QC sequencing service even when the final sequencing will be performed on a PacBio Revio. You may observe small differences in reported percent 6mA between platforms, but this is expected due to differences in sequencing chemistry, basecalling algorithms, and confidence thresholds. Despite these differences, the QC data is still useful for confirming that:

  • Labeling occurred

  • Percent 6mA is within an expected range

  • Chromatin features such as MSPs and nucleosome patterns look biologically consistent

The absolute percent 6mA may shift slightly between platforms, but the overall biological patterns should remain consistent.
