What yields are typical using this kit? What yields are required for Illumina® sequencing?

Note that EpiCypher does NOT recommend using library yields to determine assay success, as yields can vary by cell type, number of cells, target abundance, antibody quality, and assay type (please note that this kit is compatible with more than one CUTANATM assay).

The typical yield for a purified CUT&RUN sequencing library using 5 ng input DNA and following the PCR parameters in the CUT&RUN kit manual is ~300-500 ng (~30-50 ng/μL in a 10.5 μL volume; Figure 9A). Library molarity values are typically 100-200 nM, but good sequencing data can be obtained down to 0.5 nM. EpiCypher routinely uses 1-4 nM of each library for pooling, and then dilutes to the desired concentration for loading onto the sequencer.
